Title : Nonlinear Models for PET Imaging of Cigarette Smoking : Ideas for Future Application to Parkinson’s Disease and Dyskinesia
Abstract
We have developed models to describe transient effects of change in synaptic dopamine concentration on dynamic PET data (reviewed in Morris, JNM 2024). The models are collectively referred to as ‘ntPET’ (neurotransmitter PET). I will explain the evolution of these models and some variants that have been introduced by others. So far, we have applied these models to the analysis of brief dopaminergic responses to cigarette smoking and other types of drug administration (in the PET scanner). The responses have been found to be different in men and women and to reflect differences in dependence level and rates of metabolism of nicotine.
The application of models to produce time-courses of dopamine at each voxel, rather than static parameters (as produced by the standard 2-compartment model) lends itself to new approaches to data visualization. I will show “dopamine movies” of smoking.
Possible new application of models. It has been hypothesized that Tardive Dyskinesia in Parkinson’s results from long-term exposure to levodopa and a consequent disturbance of the dynamics of synthesis and release of dopamine immediately following administration of the drug (de La Fuente-Fenandez, Brain, 2004). I will discuss a possible application of the aforementioned nonlinear models to investigate this claim.
If time allows, I will review some recent findings on the effect of intense exercise on the progression of Parkinson’s disease (de Laat, NJP Parkinson’s, 2024).
